one time use of cells

I WANT TO HAVE A WORKBOOK/WORKSHEET WHERE THE USER CAN ENTER ONLY ONE TIME IN A CELL AND AFTER THAT IT GETS LOCKED ,SO THAT NO ONE ELSE CAN MODIFY THE CELL.HOW TO ACHIEVE THIS ?

How to total the visible cells?
From the first blank cell below a filtered data set, press Alt+=. Instead of SUM, you will get SUBTOTAL(9,)
See if this does what you want. Right click on your sheet tab, left click on View Code, and paste this into the module.

Modify for range of interest. Also, before you insert this code, unlock the cells in your range (in this case A1:B10), so they can accept their first and only entry.
