Is there a way to automatically populate a dropdown if that dropdown only has 1 option to select from?

So when option 1 is selected, in this example "Yes", the only available value in Option 2 is also "Yes", so instead of making a user select this can it be automatically selected?

Sure, just put an IF function in the cell. As long as the outputs don't conflict with the data validation you're golden. Try something like this: =IF(C2="Yes","Yes","")

It won't allow me to use a formula in the cell, I get the error message "A user has restricted values that can be entered into this cell".

What version of Excel are you using? I was able to do this in Excel 2007. Can you show me the exact formula you input? Are you using the List Data Validation option?
