Im trying to get the formula to:

Look in D1:Z1 for cells that match A1 then from those
Look in D2:Z2 for cells that match B1 then from that column
Look in C3:C100 for cells that match C1
Then return the value that matches all 3.

Any help appreciated.

Assuming that the values are numbers, try
Excel Formula:
=SUMIFS(INDEX(D3:Z100,MATCH(C1,C3:C100,0),0),D1:Z1,A1,D2:Z2,B1)
